چکیده

To investigate the suitable medium for in vitro shoot regeneration and plantlet growth of Dendrobium crumenatum Sw., individual protocorm-like bodies (PLBs) (about 4 - 5 mm diameter) Sw. derived from MS supplemented with 0.5 mg/L TDZ 60 days culture were cultured on 6 media; Murashige Skoog (MS) medium, 15 % (v/v) CW, CW 0.2 (w/v) AC, Vacin Went (VW) VW AC. After months culture, containing coconut water (CW) gave highest percentage shooting number shoots per explant 96.0 9.5, respectively a significant difference other media. The addition activated charcoal (AC) significantly increased leaves roots. PLBs developed into complete plantlets. AC roots root length at 5.3 34.9 mm, respectively. transfer rooted to greenhouse, 95 regenerated plantlets survived grew vigorously. Plantlets grown successfully acclimatized greenhouse showed normal development.

Multiple shoot induction from axillary bud cultures of the medicinal orchid, Dendrobium longicornu

BACKGROUND AND AIMS Dendrobium longicornu, commonly known as the 'Long-horned Dendrobium', is an endangered and medicinally important epiphytic orchid. Over-exploitation and habitat destruction seriously threaten this orchid in Northeast India. Our objective was to develop an efficient protocol for the mass propagation of D. longicornu using axillary bud segments. METHODOLOGY AND PRINCIPAL RE...

Preliminary Factors Influencing Transient Expression of Gusa in Dendrobium Savin White Protocorm-like Bodies (PLBs) Using Agrobacterium-mediated Transformation System

A critical step in the development of Agrobacterium tumefaciens-mediated transformation in orchid genus is the establishment of optimal conditions for T-DNA gene delivery into protocorm-like bodies from which the whole plantlets can be regenerated. As a first step in the development of a successful Agrobacterium tumefaciens-mediated transformation for Dendrobium savin white, factors influencing...

Genetic stability and phytochemical analysis of the in vitro regenerated plants of Dendrobium nobile Lindl., an endangered medicinal orchid

An efficient genetically stable regeneration protocol with increased phytochemical production has been established for Dendrobium nobile, a highly prized orchid for its economic and medicinal importance. Protocorm like bodies (PLBs) were induced from the pseudostem segments using thidiazuron (TDZ; 1.5 mg/l), by-passing the conventional auxin-cytokinin complement approach for plant regeneration....

Short-term storage of alginate-encapsulated protocorm-like bodies of Dendrobium nobile Lindl.: an endangered medicinal orchid from North-east India

Synthetic seed technology is an exciting and rapidly growing area of research as deals with conservation and storage of rare, endangered and desirable genotypes along with its easy handling and transportation. As propagation of many ornamental and medicinally important plant species is labour intensive, application of different growth retardants and osmotica in simple artificial seed system wou...
